TopoGEN has some exciting new products and promotions for 2017. We are radically lowering pricing on many items to kick off the new year. For example, the list price on 500 Units of Topo IIa enzyme is dropping in price from $369 to $255. Rabbit Antibody to Human Topo IIa is dropping from $329 to just $195! New products and pricing will go into effect January 1, 2017 and will be updated on our website. Why are we doing this? We have a lot of inventory we want to clear out, but more importantly we want to remain competitive in the global market place. Pricing will be good for at least the first quarter 2017. ICE BioAssay Kit (Cat. No. TG1020): ThisIn vivo Complex of Enzyme (ICE) is designed to test the ability of a drug to ‘trap’ topoisomerase I, IIa, IIb, IIIa, or IIIb on genomic DNA inside live cells. Our new kit DOES NOT require cumbersome and lengthy CsCl gradients and can be completed in a single day. Each kit includes specific antibody, control drugs (Campto or VP16) and detailed instructions. Suitable for ChIP-Seq applications, drug screening to validate in vivo poisoning activity or in vivo catalytic inhibition. Be sure to check out ICE Covalent Complex Markers to go along with the ICE Kit (Cat. No. TG1033 and TG1034).
Human Topo I High Load Kit (Cat No. TG1017): We designed this kit to facilitate detection and screening of drugs that act as Catalytic Inhibitory Compounds (CIC). In this kit, we supply exceptionally high concentration human topo I enyzme to carry out direct DNA cleavages of DNA in the absence of camptothecin. Research has shown that hu-Top1 will act semi-cooperatively at high stoichiometric ratios of enzyme:DNA. Under these conditions, topo I will introduce double strand DNA breaks in plasmid, leading to linear DNA. Strong cataltyic inhibitors will block this reaction, thereby giving scientists an additional tool to quickly find CIC agents for topo I.
Nuclear Extraction Kit (Cat. No. TG1040, TG1050). This kit provides materials and instructions for making nuclear extracts that are rich in topoisomerase activity (Top1, Top2a,2b) and low in contaminating nucleases. These extracts can be used for Western markers (using suitable antibodies), a source for topo I or II (for screening purposes) or to measure cellular topo levels in a given cell or tissue. If you are performing drug testing, this is a cheap and quick indicator of activity of a given test agent or compound. Any positives or hits, can then be validated using our purified enzymes.
Monoclonal Antibody to Human Topo I (Cat. No. TG2017). We have in beta testing form a mouse monoclonal antibody that is specific for the Top1 cleavage complex, but is unreactive to free topo I.
Purified TDP1 and TDP-2 (Cat no. TG2004H, TG2003H). We offer high purity TDP1 and TDP2, the enzymes that remove Topo from lock-up covalent complexes that form as a result of Topo poisons (VP16, camptothecin, among others). We provide validated and high specific activity enzymes for use in drug screening, mechanistic studies and R&D on enzyme action.
Non-Radioactive TDP1 and TDP2 Assay Kits (Cat. No. TG1004-1A, TG1005). TopoGEN is offering for the first time new assay kits that measure the actual release of either Top1 or Top2a,2b from covalent complexes. These do NOT require isotope or expensive oligo substrates. Assays are rapid and quantitative.
Cell Reporters to Explore Topo driven DNA damage in live cells. TopoGEN has developed a GFP reporter cell system that will reveal genotoxicity due to topo drug action. HeLa cells contain mutated GFP gene cassettes. When the GFP locus is damaged, the gene undergoes homologous recombination forming GFP positive cells. This kit is undergoing Beta testing and will be available in the 2nd quarter of 2017. It is a powerful cell based system capable of reporting events at the single cell level by time lapse immunofluorescence or by Cytofluorimetery.
Human Top1 and Top2 EMSA Kits. We will be offering a new kit to evaluate Top/DNA binding by electrophoretic shift assays in agarose gels. These kits will be released in the 2nd quarter of 2017, so please enquire.
Immunofluorescence Kit for Covalent Complexes (Top1). TopoGEN will have an IF kit to visualize covalent complexes in cells. This kit, currently in Beta testing and development will be available in Mid-2017.
Cytofluorimetry of Covalent Complex Kit. We will have a powerful new method to quantify Topo covalent complexes in live cells using Flow Cytometry. This kit will be available in Mid-2017.
